【摘要】 目的 探讨诱导型一氧化氮合酶(iNOS)、血小板衍生生长因子(PDGF)- B、脂多糖(LPS)在布-加综合征(BCS)大鼠模型中的表达及意义。方法 部分结扎大鼠肝后段下腔静脉构建BCS模型。实验分为对照组(n=20)、模型组(n=20)和假手术组(n=20),各组又分4亚组(1、3、6、12周组)。收集肝组织作免疫组织化学、苏木精-伊红和Masson染色,检测iNOS、PDGF- B和LPS表达。结果 模型组LPS值均显著高于对照组和假手术组(P=0.001)。模型组iNOS、PDGF- B mRNA和蛋白表达均显著高于对照组和假手术组(P=0.001);各亚组iNOS、PDGF- B mRNA和蛋白表达差异均有显著统计学意义(P=0.001)。模型组iNOS与PDGF- B、LPS呈正相关;肝纤维化与LPS呈正相关,与PDGF- B呈负相关。结论 BCS病损与修复是一复杂过程,iNOS、PDGF- B和LPS可能分别在BCS病程不同阶段发挥侧重不同的作用,如何调节它们在纤维化中的平衡,也许是值得进一步研究的方向。
