【摘要】 目的 探讨氧化应激(OS)在大鼠布-加综合征(BCS)肝纤维化模型中的表达及意义。方法 SD大鼠180只根据数字表法分为对照组(n=20)、模型组(n=80)和假手术组(n=80),模型组和假手术组分别均分为1、3、6、12周亚组。对照组大鼠仅常规饲养;模型组于肝后段下腔静脉(IVC)部分结扎构建BCS动物模型;假手术组未结扎IVC,其它过程同模型组。收集大鼠肝组织行苏木精-伊红(HE)、Masson和免疫组化染色,分别检测OS标志物丙二醛(MDA)、超氧化物歧化酶(SOD)与肝纤维化相关因子转化生长因子(TGF)- β1、血小板衍生生长因子(PDGF)- A表达。结果 模型组术后MDA值高于对照组,SOD值低于对照组(P均<0.05);术后6周模型组MDA表达最高,SOD表达最低。模型组TGF- β1、PDGF- A和mRNA表达高于对照组、假手术组(P均<0.05),术后3周模型组TGF- β1、PDGF- A和mRNA表达最高。模型组TGF- β1、PDGF- A与MDA互为正相关,三者均与SOD负相关。模型组肝纤维化与MDA正相关,与SOD负相关。结论 OS贯穿于BCS整个病程,影响淤血性肝纤维化进展,也许是BCS治疗切入点及未来研究方向。
