【摘要】   目的   研究深静脉血栓(DVT)大鼠模型股静脉内皮组织中转化生长因子(TGF)β1和纤溶酶原激活物抑制因子1(Serpine1)的表达变化,及在血栓形成中的作用。方法 将60只SD大鼠随机分为对照组(A组,10只)和实验组(50只)。实验组采用股静脉钳夹联合下肢石膏制动构建大鼠DVT模型。于不同时间点(创伤后2.5 h和25 h)解剖股静脉观察血栓发生率、严重程度,并将实验组分为B组(血栓形成前组,创伤后2.5 h)、C组(血栓形成组,创伤后25 h)和D组(血栓不形成组,创伤后25 h)。分离股静脉内皮组织,提取总RNA,采用Genechip Rat Genome 230 2.0基因芯片筛查差异表达的基因,进而采用实时荧光定量聚合酶链反应( PCR) 验证这些基因的表达变化,并针对上述基因进行Pathway等生物信息学分析。结果 基因芯片分析及实时PCR结果均发现创伤后2.5 h大鼠股静脉内皮组织中TGFβ1和Serpine1表达均上调,且B组高于A组和D组(P < 0.05);血栓形成时,两者亦显著上调,且 C组高于A、B、D组(P < 0.05),而A、D组间的差异无统计学意义(P > 0.05)。Pathway分析提示TGFβ1是Serpine1的上游调控基因,可诱导Serpine1的过度表达,抑制纤溶,促进血栓形成。结论 局部静脉内皮组织中TGFβ1和Serpine1表达水平上调可能在创伤性DVT形成中发挥重要作用。
