摘要:
目的评价吉西他滨对人胆管癌裸鼠移植瘤的放射增敏作用。方法建立人胆管癌裸鼠移植瘤模型,60只荷瘤鼠随机分为4组:空白对照组、单纯药物组、单纯放射治疗组、吉西他滨联合放射治疗组。单纯照射组和联合治疗组根据照射剂量不同各分为3个亚组。对照组经腹腔注射生理盐水,单纯药物组经腹腔注射吉西他滨50mg/kg;联合治疗组经腹腔注射吉西他滨50mg/kg,24h后分别进行单次10、15和20Gy的照射;隔日测量移植瘤的最大径及最小径,计算肿瘤体积及生长延缓天数等,利用增敏系数(EF)评价吉西他滨的放射增敏作用。结果单纯照射组各亚组肿瘤生长受到不同程度的抑制;联合治疗组较其他实验组肿瘤生长明显受到抑制,10、15和20Gy3个亚组的EF值分别为1.14、1.74和2.08;20Gy亚组延缓天数最长,且大于单纯药物组和单纯照射组延缓天数之和。结论照射组前24h给予吉西他滨对人胆管癌裸鼠移植瘤具有明显的放射增敏效应。
