【摘要】 目的 探讨靶向肝癌磷脂酰肌醇蛋白聚糖(GPC)-3的纳米探针Ag2S@BSA-TJ12P1在近红外二区(NIR-Ⅱ)成像早期诊断肝癌转移灶中的优势。方法 制备硫化银纳米探针Ag2S@BSA-TJ12P1。通过体外细胞实验验证GPC-3表达,检测各肝癌细胞系转移能力。采用激光共聚焦成像验证硫化银纳米探针选择性摄取至肝癌细胞系情况。采用肝癌高转移细胞系建立裸鼠肝癌原位转移模型,通过NIR-Ⅱ在体成像观察微小转移灶。结果 Ag2S@BSA-TJ12P1被激发后,可在1 080 nm处发出NIR-Ⅱ荧光。细胞实验显示,肝癌GPC-3在肝癌细胞系中均有高表达,Ag2S@BSA-TJ12P1可被HepG2、MHCC97-H、HCC-LM3细胞特异性摄取。NIR-Ⅱ成像系统对裸鼠微小转移灶作出早期诊断。 结论 Ag2S@BSA-TJ12P1可通过在体靶向肝癌GPC-3对肝癌微小转移灶成像,对肝癌微小转移灶早期诊断具有一定意义。
