【摘要】 目的 探讨一种成瘤周期短、操作便捷、安全性高、适用于肝癌治疗研究的SD大鼠原位种植型肝癌模型制作方法,同时进行彩色超声、增强CT、MR等3种影像学检查和诊断。方法 20只SD大鼠随机分为McA- RH7777组和N1S1组,每组10只。称重后将McA- RH7777和N1S1肝癌细胞分别注入两组大鼠肝左中叶内,术后连续3 d腹腔注射青霉素(20万U/只)和地塞米松(2.5 mg/只),以抗感染和免疫抑制治疗。术后7 d,采用彩色超声、增强CT、MR鉴别肝脏肿瘤,记录模型鼠体重,取出肿瘤组织进行形态学观察和病理学检查。结果 术后7 d,McA- RH7777组、N1S1组大鼠分别死亡2只、1只,影像学检查显示成活大鼠全部成瘤,建模成功率分别为80%(8/10)和90%(9/10),差异无统计学意义(P=0.86);大鼠体重分别减轻(50.00±24.93) g、(36.44±7.75) g;肿瘤体积分别为(316.43±265.84) mm3、(188.58±137.47) mm3。两组大鼠处死后,尸检结果与影像学检查一致;肉眼观察McA- RH7777肝癌外观呈团块状,N1S1肝癌呈结节状。HE染色显示McA- RH7777肝癌为高分化肝癌,N1S1肝癌为低分化肝癌。结论该方法能成功、高效地构建SD大鼠原位肝癌模型,同时通过不同影像学技术实现诊断和评价,为肿瘤介入治疗研究提供了一种可供选择的实验工具。
