摘要:
目的研究I型钙蛋白酶(calpain-I)在兔脑血管痉挛(CVS)模型中的表达,探讨钙蛋白酶在CVS中的意义。方法25只健康清洁级新西兰大白兔,5只为正常对照组,20只采用血管内穿刺法制成脑血管痉挛模型,随机分为蛛网膜下腔出血(SAH)12h、1d、3d、7d组,用免疫组化染色(SP法)观察基底动脉平滑肌中calpain-I的表达,并采用组织化学评分法进行半定量分析,观察其表达的变化。结果对照组基底动脉见极少量calpain-I阳性染色平滑肌细胞,SAH12h组见少量阳性细胞,随着时间的延长,阳性细胞数显著增多,染色强度明显增加,以SAH后3d组为最明显,在SAH后7d组仍可见大量阳性染色平滑肌细胞,但染色强度下降。半定量分析显示SAH后3d为calpain-I表达高峰,各组之间calpain-I表达差异具有统计学意义(P<0.01)。结论SAH后痉挛的脑血管平滑肌calpain-I表达增强,且具有早期、渐变、持续性的特点,calpain-I可能参与CVS血管壁结构性改变的信号调节过程。
